PCR技术是分子生物学研究和临床分子诊断的一种基本技术手段,数字PCR技术则是PCR技术和现代微机电技术相结合的产物。以PCR的理论和技术体系为基础,结合现代微机电和光学检测技术,实现单分子水平的核酸精确定量检测。数字PCR技术将DNA或RNA样品划分成单分子水平的微反应单元,然后对众多微反应单元内的靶序列进行扩增和光学检测,实现绝对定量,该技术的诞生和发展为核酸精确测定提供了一种全新的思路,并具有比传统实时荧光定量PCR技术更好的精确性和灵敏度。目前,该技术已经在肿瘤个体化诊疗、基因编辑、液体活检等生命科学研究及生物医学检测等多个领域实现应用。本书着重介绍了数字PCR技术特征、仪器系统发展、实验设计方法、试验数据处理及应用发展现状,并拓展介绍了一些实验操作和工程知识。
